Apical constriction facilitates epithelial sheet bending and invagination during morphogenesis. Apical constriction is conventionally thought to be driven by the continuous purse-string-like contraction of a circumferential actin and non-muscle myosin-II (myosin) belt underlying adherens junctions. However, it is unclear whether other force-generating mechanisms can drive this process. Here we show, with the use of real-time imaging and quantitative image analysis of Drosophila gastrulation, that the apical constriction of ventral furrow cells is pulsed. Repeated constrictions, which are asynchronous between neighbouring cells, are interrupted by pauses in which the constricted state of the cell apex is maintained. In contrast to the purse-string model, constriction pulses are powered by actin-myosin network contractions that occur at the medial apical cortex and pull discrete adherens junction sites inwards. The transcription factors Twist and Snail differentially regulate pulsed constriction. Expression of snail initiates actin-myosin network contractions, whereas expression of twist stabilizes the constricted state of the cell apex. Our results suggest a new model for apical constriction in which a cortical actin-myosin cytoskeleton functions as a developmentally controlled subcellular ratchet to reduce apical area incrementally.

Hello my computer keeps freezing, all of a sudden at most random times. I have tried reinstalling windows, going back to older drivers, and defaulting bios settings. When i enter the Event Viewer it shows the events as 10110 and 10111.

Hi Natalia!I have a problem with the way google sheets interprets the dates I'm writting. I have a column for dates with the format dd-mm-yyyy. However, when I type a date such as 12-2-23 it assumes the date 23-12-2012 by default! What can I do? Thanks!

As i am trying to use countifs formula for eg:- column C has filled with dark and column b with date , so I want to use this formula in a way if i give a date range for 15th jan to 20th jan how many count of dark is there while trying a formula in excel it was working but not in google sheet as there was some issue with date format while trying to change the date format for few cells it got changed but for few it didnt how to resolve these both issues

Hi i have 1 problem with dates if i type 23/8 it must put in the date 23/8/2020 according to my farnatting but it doesnt it only show 23/8 as data and not a date because it reads the 23 as month if i type 8/23 then it show the date 23/8/2020. I need to type the month first and then the date for google sheets to read it as a date. If i type the date and then the month as with excel, google sheets does not read it as a date. I'm noticing an issue where I'm getting partially uploaded/corrupted files in google Photos. I'm thinking that drive is picking up and starting to sync the files before they have actually finished copying with rclone.

The problem I'm trying to solve is that I don't think rclone is LOCKING the file while its copying data - hence google drive picks it up and starts uploading it before its complete. This results in a corrupted/partial file ending up in google photos.

so the next question, why try to do that?--- to use the same directory as a source with gdrive sync progream.--- to use the same directory as a dest with rclone--- to allow both programs to sync at the same time

from what i have read, that google sync app has built-in support to pause syncing,i find it hard to believe that the app cannot handle pausing gracefully.or just quit that app using its user interface
